Summary: Burnout Syndrome is an occupational condition related to chronic work-related stress that, although it is a frequent cause of work leave, is not classified as a medical condition by the World Health Organization, nor has it established definitive diagnostic criteria. Therefore, data on its incidence may not accurately reflect the true prevalence of cases. This context highlights the importance of studies employing screening methods based on biological markers, which may contribute to greater diagnostic robustness. From this perspective, BDNF gene methylation has emerged as an important candidate due to its responsiveness to environmental stimuli, as well as its sensitivity to epigenetic alterations mediated by environmental factors. Accordingly, the present study aimed to investigate the association between Burnout symptoms and methylation of the exon IV promoter of the BDNF gene. This study was conducted as part of the SOMA-SI Project, which included public security professionals from the state of Espírito Santo, Brazil, in 2023. Participants were initially screened for perceived stress and subsequently assessed for symptoms of Burnout, depression, anxiety, and the use of antidepressant medications. To evaluate DNA methylation, blood samples were collected, followed by DNA extraction and subsequent pyrosequencing. The data were organized, filtered, and subjected to statistical analysis. The results revealed that more than 70% of participants presented symptoms of Burnout. An inverse relationship was observed between Burnout symptoms and methylation levels at CpG sites 5 and 7. Additionally, the results showed that women presented higher levels of Burnout symptoms compared to men, as well as a greater occurrence of concomitant psychological conditions. This study reveals a distinct methylation profile among public security professionals, highlighting the relevance of the topic and the need for future longitudinal studies.
